Non-invasive method for the evaluation of IL-6 and IL-10 levels in patients with chronic hepatitis C

چکیده
With over 180 million people affected worldwide, hepatitis C is considered a global problem. In order to establish chronic infections, the virus interferes with cellular mechanisms coordinated by cytokines’ network. Chronic HCV infection is associated with oral manifestations including periodontal disease. The aim of this study was to evaluate the levels of IL-6 and IL-10 in gingival crevicular fluid (GCF) versus plasma, in patients with chronic hepatitis C, in order to estimate the potential of oral fluids as substitute of blood for the evaluation of immune responses in this disease. Material and methods. Sample-pairs consisted of blood and GCF harvested from 32 anti-HCV positive patients (age range 35-74 y.o) and from control group (10 subjects without HCV infection), which were tested for anti-HCV antibodies, HCV viral loads and IL-6 and IL-10 levels. The dental evaluation was based on CPITN criteria. Results. All investigated patients were anti-HCV positive in plasma and GCF and HCV RNA was detected in 20/32 sample pairs. Significant differences were observed for IL-10 in plasma versus GCF (p=0.0025), but not for IL-6 (p= 0.44). The expression of IL-10 in GCF displays a significant correlation with the CPITN index. Conclusions. Elevated GCF levels of IL10 correlated with higher CPITN index and viremia, accounting for usefulness of GCF as an alternative non-invasive source for IL-10 detection in HCV positive cases.

Comparison between interleukine-17 levels by NK-T cells in patients with chronic hepatitis C and healthy individuals
Background and Aims: Natural killer T (NKT) cells have been suggested to play critical roles in a wide range of immune responses especially against hepatotropic pathogens such as hepatitis C virus (HCV). In the present study, we investigated the status of NKT cells by producing interleukin-17 cytokine in peripheral bl...
متن کاملImmunogenicity Evaluation of a DNA Vaccine Expressing the Hepatitis C Virus Non-Structural Protein 2 Gene in C57BL/6 Mice
Backgrounds: Most of the hepatitis C virus (HCV) infections elicit poor immune responses and 75% to 85% of cases become chronic therefore, the development of an effective vaccine against HCV is of paramount importance. In this study, we aimed to evaluate co-administration of HCV non-Structural Protein 2 and IL-12 DNA vaccines in C57BL/6 mice. Methods: A plasmid encoding full-length HCV NS2 prot...

Background and purpose: Host genetic and environmental factors and factors associated with hepatitis C virus (HCV) play critical roles in development of the hepatitis. This study was performed to investigate the association between Interleukin-28B (IL-28B) genetic variants and chronic hepatitis C. Materials and methods: This case-control study was conducted in 133 HCV-RNA positive patients att...
متن کاملEffects of Non-Surgical Periodontal Therapy on Gingival Crevicular Fluid Levels of Interleukin-17 and Interleukin-23 in Patients with Periodontitis: A Clinical Trial
Background and Aim: Considering the critical role of inflammatory responses in periodontal disease, knowledge about the immune system and cytokines seems nec-essary. The aim of this study was to evaluate the level of IL-23 and IL-17 in gingival crevicular fluid (GCF) of patients with chronic and aggressive periodontitis, before and after non-surgical periodontal therapy. Materials and Methods:...
متن کاملبر IL 28B ژن rs اثر پلی مورفیسم تک نوکلئوتیدی 12979860 پاسخ به درمان با پگ- اینترفرون/ریباورین در بیماران C ایرانی مبتلا به هپاتیت
Background and purpose: The standard of care treatment for infected patients with HCV is based on a combination of pegylated interferon alpha and ribavirin. Recently, the rs12979860 SNP polymorphism, located upstream of the interleukin 28B gene, was shown to be strongly associated with response to anti-HCV therapy. This study investigated the distribution of the (C/T) polymorphism with sust...
